This chapter describes the rational design and characterization of Dimeric-DesAb-O, engineered to target Aβ42 oligomers. Real-Time Based ELISA assay and Thioflavin T (ThT) assays confirm that dimerization enhances binding affinity and slows Aβ42 aggregation, while Transmission Electron Microscopy (TEM) shows reduced fibril diameter. Dimeric-DesAb-O selectively detects oligomers in AD patient CSF and neutralizes neurotoxicity more effectively than the monomer. These results prove that rational dimerization improves target engagement, offering a potent strategy for AD diagnosis and therapy.
