This chapter describes the materials and methods employed across the three main sections of the study. It details patient recruitment and neuropsychological examination, CSF collection, and biochemical assays such as circular dichroism (CD), dynamic light scattering (DLS), and intrinsic tryptophan fluorescence. The methodological framework also encompasses cell culture models, intracellular calcium measurements, MTT reduction assays, and advanced imaging techniques like STED microscopy. Furthermore, it outlines the purification of DesAb-O and Dimeric-DesAb-O constructs, including the characterization of their binding properties through dot blot analysis and Real-Time based ELISA assay.
